細胞因子 ELISA 板陣列
細胞因子和生長因子是在許多生物過程中起關鍵作用的信號分子，如細胞生長、分化、基因表達、遷移、免疫和炎癥。這些信號的破壞會導致多種疾病，包括關節炎、肝病、炎癥性腸病、心臟相關疾病和癌癥。
Signosis提供了許多工具來分析一種特定的細胞因子或生長因子，或同時監測多種細胞因子或生長因子。
原則
在該測定中，測試樣品最初與固相捕獲抗體反應，導致細胞因子與孔結合。然后洗滌孔以去除未結合的蛋白質，并加入生物素連接的抗體以與細胞因子結合。洗去未結合的抗體后，加入鏈霉親和素-HRP偶聯物，與抗體結合的細胞因子形成復合物。
對于比色試劑盒，添加 HRP 底物 TMB，并在檢測到 HRP 連接的抗體時形成藍色。然后用終止溶液終止反應，終止溶液將顏色從藍色變為黃色。每個孔中的細胞因子濃度與其顏色強度成正比，可以通過在酶標儀中測量其 450 nm 處的光密度 （OD450） 來量化。
對于化學發光試劑盒，用HRP發光底物檢測板。在微孔板發光儀上，發光以相對光單位 （RLU） 的形式報告。每種特定細胞因子的表達水平與發光強度成正比。
miRNA實時熒光定量PCR
使用 miRNA 實時熒光定量 PCR 試劑盒，可以從總 RNA 或直接從細胞裂解物中分析 miRNA，無需 cDNA 轉化。該試劑盒可用于 12 個樣品和 50 個 PCR 反應。
原則
在測定中，靶 miRNA 分子與兩個寡核苷酸雜交形成 RNA/DNA 雙鏈體。當序列wan全匹配時，它們與 miRNA 對齊，并且關節可以與 DNA 連接酶連接。miRNA之間的單核苷酸差異將阻斷雜交或連接。連接一對寡核苷酸后，對連接的分子進行實時熒光定量PCR分析。
